A den is what people say when the room isn’t a “legal” bedroom. In order to be legal, a bedroom needs to have a second means of egress — either a window or door to the exterior. The window must meet certain size requirements for egress, must open a certain amount, and the window sill must be no more than 44″ above the floor — basically, accessible enough for someone to crawl out and for a fire fighter to be able to crawl in, if necessary.
Sometimes bedrooms have windows, but if they don’t meet those particular requirements it must still be counted as a den, not a bedroom.
